RESUMO

Leopoldia comosa (LC), popularly known as Muscari comosum, spontaneously grows in the Mediterranean region and its bulbs are used as a vegetable. Traditionally, they are also used to treat various diseases and conditions, which has inspired the study of the pharmacological activities of different parts of LC. These studies revealed the numerous biological properties of LC including antioxidant, anti-inflammatory, anti-diabetes, anti-obesity, anti-cancer, anti-Alzheimer's disease, antibacterial, and immune stimulant. High antioxidant activity compared to other non-cultivated plants, and the potential role of antioxidant activity in other reported activities make LC an excellent candidate to be developed as an antioxidant plant against important associated diseases. The presence of a diverse class of phytochemicals (n = 85), especially flavonoids and homoisoflavones, in LC, also imparts significance to the nutraceutical candidature of the plant. However, limited animal studies and the lack of a directional approach have limited the further design of effective clinical studies for the development of LC. The current study is the first attempt to comprehensively compile information regarding the phytochemicals and pharmacological activities of LC, emphasize the targets/markers targeted by LC, important in other activities, and also highlight the current gaps and propose possible bridges for the development of LC as a therapeutic and/or supplement against important diseases.

Muscaris is a modern white grape variety with good fungal resistance and a pleasant aroma, the molecular background of which was unknown. A comparative aroma extract dilution analysis applied to Muscaris grapes and grapes of the father variety Muskateller revealed little differences and resulted in 39 and 35 odorants, respectively. Sixteen odorants exceeded their odor threshold concentrations. Odor reconstitution and omission experiments showed that the distinct lychee note in the aroma of the Muscaris grapes was generated by the combination of (2S,4R)-rose oxide and geraniol. This finding will guide further molecular research on the transfer of the lychee note into wine and may also be helpful for the targeted breeding of new grape varieties.

Since ancient times, Polygonatum Mill. (Asparagaceae) has been utilized as a medicinal and culinary resource in China. Its efficacy in treating various illnesses has been well documented. Traditional processing involves the Nine-Steam-Nine-Bask method, which results in a reduction of toxicity and enhanced effectiveness of Polygonatum. Many substances, such as polysaccharides, lignins, saponins, homoisoflavones, alkaloids, and others, have been successfully isolated from Polygonatum. This review presents the research progress on the chemical composition of three crude and processed Polygonatum, including Polygonatum sibiricum Redouté (P. sibiricum), Polygonatum kingianum Collett & Hemsl (P. kingianum), and Polygonatum cyrtonema Hua (P. cyrtonema). The review also includes the pharmacology of Polygonatum, specifically on the pharmacology of polysaccharides both before and after processing. Its objective is to provide a foundation for uncovering the significance of the processing procedure, and to facilitate the development and utilization of Polygonatum in clinical practice.

Polygonatum is the largest genus of tribe Polygonateae (Asparagaceae) and is widely distributed in the temperate Northern Hemisphere, especially well diversified in southwestern China to northeastern Asia. Phylogenetic relationships of many species are still controversial. Hence it is necessary to clarify their phylogenetic relationships and infer possible reticulate relationships for the genus. In this study, genome-wide data of 43 species from Polygonatum and its closely related taxa were obtained by Hyb-Seq sequencing. The phylogenetic trees constructed from genome-wide nuclear and chloroplast sequences strongly supported the monophyly of Polygonatum with division into three major clades. A high level of incongruence was detected between nuclear and chloroplast trees as well as among gene trees within the genus, but all occurred within each major clade. However, introgression tests and reticulate evolution analyses revealed low level of gene flow and weak introgression events in the genus, suggesting hybridization and introgression were not dominant during the evolutionary diversification of Polygonatum in the Northern Hemisphere. This study provides important insights into reconstructing evolutionary relationships and speciation pattern of taxa from the north temperate flora.

The rhizome of Rohdea chinensis (Baker) N.Tanaka (RRc) is a famous folk medicine for the treatment of carbuncles and pharyngitis. Steroidal saponins (SSs) were considered to be the most abundant active constituents in RRc. However, to date, the in-depth study of SSs is still lacking. This study was aimed to investigate the SSs profiles of RRc extract by HPLC-ESI-QTOF-MS/MS. Analysis was performed on an Agilent poroshell 120 EC-C18 column (2.1 mm × 100 mm, i.d., 2.7 µm) with 0.1% formic acid aqueous solution and acetonitrile as the mobile phase under gradient conditions. The results showed that 32 SSs including 20 furospirostanol, 11 spirostanol and 1 pseudo-spirostanol saponins were identified, 5 of which were reported in this plant for the first time. This is the first report on the analysis of SSs in RRc. This novel analysis method may stimulate further research regarding the identification of SSs in other plant species.

Indoor low-concentration carbon monoxide (CO) exposure is widespread worldwide, and potted plants may be a potential means for CO purification. The objective is to evaluate common indoor plants' CO purification and tolerance capacities. Epipremnum aureum (Linden ex André) G.S.Bunting, Chlorophytum comosum (Thunb.) Jacques, Spathiphyllum kochii Engl. & K.Krause, and Sansevieria trifasciata Hort. ex Prain with similar sizes were tested in the glass chamber with initial CO concentrations of 10, 25, 50, 100, 200, and 400 ppm, respectively. (1) The CO purification capacity of the four potted plants is ranked as Epipremnum aureum (Linden ex André) G.S.Bunting > Chlorophytum comosum (Thunb.) Jacques > Spathiphyllum kochii Engl. & K.Krause > Sansevieria trifasciata Hort. ex Prain. Under the purification effect of each plant, the CO concentration in the chamber decreases linearly and significantly (p < 0.05), and within a specific time period, the time-weighted average (TWA) CO concentrations can be reduced to below the corresponding permissible exposure limits specified by some countries and organizations. (2) With the increase of the stomatal number of each plant and the increase in CO concentration, the hourly and cumulative absorbed CO of each plant increase linearly and significantly (p < 0.05). (3) With the increase in CO concentration, the CO purification efficiency of each plant decreases exponentially and significantly (p < 0.05). (4) When the CO concentration was ≤ 50 ppm, all plants could effectively purify CO without damage. When the CO concentration was in the range of 100 ~ 400 ppm, within 2 weeks after the 48-h experiment, the leaf tips of Chlorophytum comosum (Thunb.) Jacques and Epipremnum aureum (Linden ex André) G.S.Bunting were damaged one after another, and the damaged leaf area increased with the increase of CO concentration. However, each plant as a whole still survived. This study demonstrated that different species of potted plants can effectively absorb low concentrations of CO to varying degrees, but higher concentrations of CO will damage the survival of specific species of potted plants.

This study aims to formulate bacterial tablets of cadmium (Cd)-resistant Micrococcus sp. MU1, an indole-3-acetic acid-producer, for soil inoculation to improve Cd phytoremediation by Chlorophytum comosum (Thunb.) Jacques. The viability of Micrococcus sp. MU1 in tablets after storage at room temperature and 4 °C was determined. The ability of Micrococcus sp. tablets and cell suspensions on stimulating growth and Cd accumulation in C. comosum was compared. The results found that the viability of Micrococcus sp. tablets stored at room temperature and 4 °C for 2 months were 29.2 and 97.9%, respectively. After 2 months of growth in pots, the dry biomass weights of C. comosum amended with Micrococcus sp. tablet and cell suspension were greater than that of uninoculated control by 1.4- and 1.3-fold, respectively. Cd concentrations in the roots and shoots of C. comosum inoculated with bacterial tablet and bacterial suspension were not significantly different (p < 0.05) and were greater than that of the uninoculated plants. In addition, plants inoculated with Micrococcus sp. tablet and cell suspension exhibited superior phytoextraction performance, bioaccumulation factor, and translocation factor, indicating equal performance of both bacterial forms on boosting Cd phytoremediation efficiency in C. comosum. These findings suggest that soil inoculation with Micrococcus sp. tablet as a ready-to-use inoculum is a novel approach to promote phytoremediation of C. comosum in Cd-contaminated agricultural soil.

BACKGROUND: Chromosome number and genome size changes via dysploidy and polyploidy accompany plant diversification and speciation. Such changes often impact also morphological characters. An excellent system to address the questions of how extensive and structured chromosomal changes within one species complex affect the phenotype is the monocot species complex of Barnardia japonica. This taxon contains two well established and distinct diploid cytotypes differing in base chromosome numbers (AA: x = 8, BB: x = 9) and their allopolyploid derivatives on several ploidy levels (from 3x to 6x). This extensive and structured genomic variation, however, is not mirrored by gross morphological differentiation. RESULTS: The current study aims to analyze the correlations between the changes of chromosome numbers and genome sizes with palynological and leaf micromorphological characters in diploids and selected allopolyploids of the B. japonica complex. The chromosome numbers varied from 2n = 16 and 18 (2n = 25 with the presence of supernumerary B chromosomes), and from 2n = 26 to 51 in polyploids on four different ploidy levels (3x, 4x, 5x, and 6x). Despite additive chromosome numbers compared to diploid parental cytotypes, all polyploid cytotypes have experienced genome downsizing. Analyses of leaf micromorphological characters did not reveal any diagnostic traits that could be specifically assigned to individual cytotypes. The variation of pollen grain sizes correlated positively with ploidy levels. CONCLUSIONS: This study clearly demonstrates that karyotype and genome size differentiation does not have to be correlated with morphological differentiation of cytotypes.

One of the central goals of evolutionary biology is to understand the genomic basis of adaptive divergence. Different aspects of evolutionary processes should be studied through genome-wide approaches, therefore maximizing the investigated genomic space. However, in-depth genome-scale analyses often are restricted to a model or economically important species and their closely related wild congeners with available reference genomes. Here, we present the high-quality chromosome-level genome assembly of Chouardia litardierei, a plant species with exceptional ecological plasticity. By combining PacBio and Hi-C sequencing technologies, we generated a 3.7 Gbp genome with a scaffold N50 size of 210 Mbp. Over 80% of the genome comprised repetitive elements, among which the LTR retrotransposons prevailed. Approximately 86% of the 27,257 predicted genes were functionally annotated using public databases. For the comparative analysis of different ecotypes' genomes, the whole-genome sequencing of two individuals, each from a distinct ecotype, was performed. The detected above-average SNP density within coding regions suggests increased adaptive divergence-related mutation rates, therefore confirming the assumed divergence processes within the group. The constructed genome presents an invaluable resource for future research activities oriented toward the investigation of the genetics underlying the adaptive divergence that is likely unfolding among the studied species' ecotypes.

Polygonatum Miller belongs to the tribe Polygonateae of Asparagaceae. The horizontal creeping fleshy roots of several species in this genus serve as traditional Chinese medicine. Previous studies have mainly reported the size and gene contents of the plastomes, with little information on the comparative analysis of the plastid genomes of this genus. Additionally, there are still some species whose chloroplast genome information has not been reported. In this study, the complete plastomes of six Polygonatum were sequenced and assembled, among them, the chloroplast genome of P. campanulatum was reported for the first time. Comparative and phylogenetic analyses were then conducted with the published plastomes of three related species. Results indicated that the whole plastome length of the Polygonatum species ranged from 154,564 bp (P. multiflorum) to 156,028 bp (P. stenophyllum) having a quadripartite structure of LSC and SSC separated by two IR regions. A total of 113 unique genes were detected in each of the species. Comparative analysis revealed that gene content and total GC content in these species were highly identical. No significant contraction or expansion was observed in the IR boundaries among all the species except P. sibiricum1, in which the rps19 gene was pseudogenized owing to incomplete duplication. Abundant long dispersed repeats and SSRs were detected in each genome. There were five remarkably variable regions and 14 positively selected genes were identified among Polygonatum and Heteropolygonatum. Phylogenetic results based on chloroplast genome strongly supported the placement of P. campanulatum with alternate leaves in sect. Verticillata, a group characterized by whorled leaves. Moreover, P. verticillatum and P. cyrtonema were displayed as paraphyletic. This study revealed that the characters of plastomes in Polygonatum and Heteropolygonatum maintained a high degree of similarity. Five highly variable regions were found to be potential specific DNA barcodes in Polygonatum. Phylogenetic results suggested that leaf arrangement was not suitable as a basis for delimitation of subgeneric groups in Polygonatum and the definitions of P. cyrtonema and P. verticillatum require further study.

Crassulacean acid metabolism - or CAM photosynthesis - was described in the early to mid-20th century, and our understanding of this metabolic pathway was later expanded upon through detailed biochemical analyses of carbon balance. Soon after, scientists began to study the ecophysiological implications of CAM, and a large part of this early work was conducted in the genus Agave, in the subfamily Agavoideae of the family Asparagaceae. Today, the Agavoideae continues to be important for the study of CAM photosynthesis, from the ecophysiology of CAM species, to the evolution of the CAM phenotype and to the genomics underlying CAM traits. Here we review past and current work on CAM in the Agavoideae, in particular highlighting the work of Park Nobel in Agave, and focusing on the powerful comparative system the Agavoideae has become for studying the origins of CAM. We also highlight new genomics research and the potential for studying intraspecific variation within species of the Agavoideae, particularly species in the genus Yucca. The Agavoideae has served as an important model clade for CAM research for decades, and undoubtedly will continue to help push our understanding of CAM biology and evolution in the future.

RESUMO

The phytochemical constituent investigation on the 70 % ethanol extract of the rhizomes of Tupistra chinensis Baker resulted in the isolation of three new steroidal saponins which were named tuchinosides A-C (1-3). Their structures were determined by extensive spectrum analysis and chemical evidence, especially 2D NMR and HR-ESI-MS techniques. In addition, the cytotoxicity of compounds 1-3 against several human cancer cell lines was evaluated.

Grape hyacinth (Muscari spp.) is a famous bulbous blue flower; however, few bicolor varieties are available in the market. Therefore, the discovery of bicolor varieties and understanding of their mechanisms are crucial to the breeding of new varieties. In this study, we report a significant bicolor mutant with white upper and violet lower portions, with both parts belonging to a single raceme. Ionomics showed that pH and metal element contents were not responsible for the bicolor formation. Targeted metabolomics illustrated that the content of the 24 color-related compounds was significantly lower in the upper part than that in the lower part. Moreover, full-length transcriptomics combined with second-generation transcriptomics revealed 12,237 differentially expressed genes in which anthocyanin synthesis gene expression of the upper part was noted to be significantly lower than that of the lower part. Transcription factor differential expression analysis was used to describe the presence of a pair of MaMYB113a/b sequences, with low levels of expression in the upper part and high expression in the lower part. Furthermore, tobacco transformation confirmed that overexpression of MaMYB113a/b can promote anthocyanin accumulation in tobacco leaves. Accordingly, the differential expression of MaMYB113a/b contributes the formation of a bicolor mutant in Muscari latifolium.

In indoor environments, cooking is a major contributor to indoor air pollution releasing potentially harmful toxic compounds such as polycyclic aromatic hydrocarbons. In our study, Chlorophytum comosum 'Variegata' plants were applied to monitor PAH emission rates and patterns in previously selected rural Hungarian kitchens. Concentration and profile of accumulated PAHs could be well explained by cooking methods and materials used in each kitchen. Accumulation of 6-ring PAHs was characteristic in the only kitchen which frequently used deep frying. It also should be emphasized that applicability of C. comosum as indoor biomonitor was assessed. The plant has proven a good monitor organism as it accumulated both LMW and HMW PAHs.

For centuries, humans occupying arid regions of North America have maintained an intricate relationship with Agave (Agavoideae, Asparagaceae). Today Agave cultivation, primarily for beverage production, provides an economic engine for rural communities throughout Mexico. Among known dryland-farming methods, the use of rock piles and cattle-grazed areas stand out as promising approaches for Agave cultivation. Identifying new cultivation areas to apply these approaches in Arizona, USA and Sonora, Mexico warrants a geographic assessment of areas outside the known ranges of rock piles and grasslands. The objective of this study was to predict areas for dryland-farming of Agave and develop models to identify potential areas for Agave cultivation. We used maximum entropy (MaxEnt) ecological-niche-modeling algorithms to predict suitable areas for Agave dryland farming. The model was parameterized using occurrence records of Hohokam rock piles in Arizona and grassland fields cultivated with Agave in Sonora. Ten environmental-predictor variables were used in the model, downloaded from the WorldClim 2 climate database. The model identified potential locations for using rock piles as dryland-farming methods from south-central Arizona to northwestern Sonora. The Agave-grassland model indicated that regions from central to southern Sonora have the highest potential for cultivation of Agave, particularly for the species Agave angustifolia. Results suggest that there are many suitable areas where rock piles can be used to cultivate Agave in the Sonoran Desert, particularly in the border of southeastern Arizona and northwest Sonora. Likewise, cattle-grazing grasslands provide a viable environment for cultivating Agave in southern Sonora, where the expanding bacanora-beverage industry continues to grow and where different Agave products (e.g., syrups, fructans, saponins, and medicinal compounds) can potentially strengthen local economies.

Lung cancer is one of the leading cancers and causes of cancer-related deaths worldwide. Due to its high prevalence and mortality rate, its clinical management remains a significant challenge. Previously, the in vitro anticancer activity of Aspiletrein A, a steroid and a saponin from Aspidistra letreae, against non-small cell lung cancer (NSCLC) cells was reported. However, the anticancer molecular mechanism of other Aspiletreins from A. letreae remains unknown. Using in silico network pharmacology approaches, the targets of Aspiletreins were predicted using the Swiss Target Prediction database. In addition, key mediators in NSCLC were obtained from the Genetic databases. The compound-target interacting networks were constructed using the STRING database and Cytoscape, uncovering potential targets, including STAT3, VEGFA, HSP90AA1, FGF2, and IL2. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis demonstrated that several pathways were highly relevant to cancer pathogenesis. Additionally, molecular docking and molecular dynamic analyses revealed the interaction between key identified targets and Aspiletreins, including hydrogen bonding and Van der Waals interaction. This study provides potential targets of Aspiletreins in NSCLC, and its approach of integrating network pharmacology, bioinformatics, and molecular docking is a powerful tool for investigating the mechanism of new drug targets on a specific disease.

The search for therapeutic agents that improve kidney function against doxorubicin-induced renal toxicity is important. Herein, the potential nephroprotective activity by Asparagus falcatus L. (AF, Asparagaceae) leaf extracts against doxorubicin-induced renal toxicity (5 mg/kg, ip) in Wistar rats (n = 6/group) after oral administration of hexane (55 mg/kg), ethyl acetate (35 mg/kg), butanol (75 mg/kg), and aqueous (200 mg/kg) extracts of AF for 28 consecutive days was investigated. It was noticed that the treatment with the selected extracts of AF significantly attenuated doxorubicin-induced elevations of serum creatinine, urea nitrogen, ß2-microglobulin, cystatin C, and proteinuria in experimental rats. The histology showed attenuation of the features of acute tubular injury. Treatment regimens significantly reversed the doxorubicin-induced reduction in total antioxidant status, glutathione peroxidase, and glutathione reductase activity in renal tissue homogenates. A suppression in lipid peroxidation was noted with hexane, ethyl acetate, and butanol extracts of AF. Moreover, a reduction in the concentration of the pro-inflammatory mediator TNF-α (p < 0.05), and immunohistochemical expression of COX-2 were observed. The immunohistochemical expression of pro-apoptotic Bax protein was decreased and the anti-apoptotic BCL-2 was increased in renal tissues following the treatments. In conclusion, it was revealed that, hexane, ethyl acetate, butanol, and aqueous extracts of AF attenuate doxorubicin-induced renal toxicity in Wistar rats through antioxidant, anti-inflammatory, and anti-apoptotic pathways. The plant, AF could be recommended as a promising therapeutic agent to minimize renal toxicity induced by doxorubicin in cancer patients, however, subsequent clinical trials are warranted.

Scilla siberica subsp. armena is known as Siberian Squill which is naturally distributed in Lebanon-Syria, Transcaucasus, and Turkey. It is a perennial bulbous plant with a short vegetation period from March to May. In this study, the antimicrobial, antioxidant, α-amylase, and α-glucosidase inhibitory activities of corm, leaf, and flower methanolic extracts were examined. The anatomy and morphology of the plant organs were investigated by light and electron microscopes. The anatomy of S. siberica subsp. armena was investigated for the first time by electron microscopy in this study. The corm, leaf, and flower methanolic extracts were assessed against E. coli ATCC 8739, S. aureus ATCC 6538, B. subtilis ATCC 19,659, C. albicans ATCC 10,231, C. krusei ATCC 14,243, and C. tropicalis ATCC 750. Among the extracts, that obtained from the blue pollen showed the best antimicrobial activity against C. tropicalis ATCC 750 strain with a MIC value of 312.5 µg/mL. The highest phenolic content was determined in leaf extract with 53.59211 µg GAE/mg extract value. The extract showed the best anti-lipid peroxidation activity with 376.69 µg/mL value. Using DPPH· and ABTS·+ tests, it was determined that the flower and leaf extracts have the best activity (IC50 = 756.13 µg/mL and IC50 = 94.07 µg/mL, respectively). The flower extract exhibited α-glucosidase inhibitory activity with the IC50 value of 5239 µg/mL. Based on the presented results of the in vitro antimicrobial, antioxidant, and antidiabetic activities of the S. siberica subsp. armena, we suggest that natural compounds from S. siberica subsp. armena are of potential use for the improvement of an antidiabetic, antioxidant, and antimicrobial agent.

PURPOSE: This experiment aimed to investigate the effect of gamma irradiation on morpho-physiological characteristics and molecular-induced variations in Polianthes tuberosa L. METHODS: Experiments were designed according to a completely randomized design with eight different gamma-ray doses (0, 20, 30, 40, 50, 60, 70, and 80 Gy) via a source of cobalt-60 with three replications. Some morpho-physiological characteristics of tuberoses were screened and evaluated at the end of the flower growth and development phases. The RAPD-PCR molecular marker technique was further used to identify the mutants of phenotypic variation flowers. RESULTS: Results indicated that the effect of different levels of γ-rays on some morphological and physiological traits was significant as the gamma-ray level was increased up to 50 Gy. The doses higher than 50 Gy were found to cause stand or no growth. The 50 Gy gamma irradiation reduced germination by 70.59%, germination rate by 66.36%, dry weight by 88.15%, fresh weight by 87.41%, flowering stem height (cm) by 69.22%, leaf area (cm2) by 57.35%, leaf number by 34.41%, chlorophyll content (mg g-1 FW) by 44.79%, number of florets by 92.57%, spike height (cm) by 27.80%, bulblet number by 32.57%, and bulblet diameter (mm) by 30.21%. On the contrary, gamma radiation at 50 Gy increased relative water content (%) and electrolyte leakage (ds m-1) by 41.27 and 237.65%, respectively. The results also showed that bulbs treated with 20 Gy gamma ray had the highest germination percentage and dry weight. The RAPD analysis indicated that among 10 primers tested, nine primers showed clear bands as the highest number of amplified fragments (90) was related to the OPM13 primer and the lowest number (40) to the OPM10 primer. However, the DNA polymorphism was dose-dependent. CONCLUSION: Overall results showed that although the plant morphology was changed with gamma-ray level, no changes occurred in tuberose color.

R2R3 MYBs play vital roles in the regulation of flavonoid biosynthesis. However, the regulatory network of R2R3 MYBs in flavonoid biosynthesis is not fully understood in grape hyacinth (Muscari spp.). Here, we identified two R2R3 MYBs, MaMYBF and MaMYB1, as potential regulators of flavonol and anthocyanin biosynthesis, respectively. MaMYBF and MaMYB1 expression was elevated during flower development and was light-induced, and the expression patterns were related to those of the flavonoid structural genes MaFLS and MaDFR, respectively. The BiFC assay verified that MaMYB1 interacts with MabHLH1, but MaMYBF does not. A dual luciferase assay revealed that MaMYBF alone strongly activated pMaFLS, and its activation was attenuated at reduced doses of MaMYBF in the presence of MabHLH1, MaMybA, and MaMYB1. MaDFR transcription mediated by MaMybA and MabHLH1 was inhibited by MaMYB1. Moreover, overexpression of MaMYBF and MaMYB1 in tobacco reduced flower pigmentation and repressed the expression of flavonoid pathway key structural genes. Therefore, MaMYBF regulates the flavonol pathway independently of cofactors. Whereas MaMYB1 regulates anthocyanin biosynthesis by binding to MabHLH1 and disrupting the MaMybA-bHLH complex in grape hyacinth. Our results offer new insights into the intricate regulatory network of flavonoids in grape hyacinth involving the regulation of both flavonol and anthocyanin.
